Abstract	Microglia are a bone marrow derived population of immune effector cells resident in the CNS. The function of these cells is manifold, and includes physiological, reparative, and immunological activities. During previous investigations of these cells, a panel of monoclonal antibodies was generated by immunization of mice with viable, cultured rat microglia. Bearing the moniker TLD , these antibodies recognize various antigens expressed by microglia. Upon determination of the specificity of various TLD antibodies, one clone, TLD-1A8A, proved to be specific for Antisecretory Factor (ASF). Not previously known to be expressed by microglia, ASF had been defined as a protein which counteracted enterotoxin induced intestinal fluid secretion. This thesis documents the investigations of the immunological function of ASF. ASF is expressed by members of the monocyte/macrophage/microglial group, and to some extent by B cells, but not by T cells. ASF expression by cells that morphologically appear to be macrophages and dendritic cells can be demonstrated by immunohistochemistry in the spleen, lymph node, thymus, and Peyer's patch. ASF is also strongly expressed by perivascular cells in the CNS. The distribution and expression levels of ASF are markedly altered during an immunological response. In vitro , the expression levels of ASF can be modified by IFN- ?, LPS, and TNF- ?. Immunological function of ASF can be demonstrated both in vitro and in vivo. In vitro , TLD-1A8A has the ability to increase T cell proliferation and alter cytokine profiles in T cell proliferation assays. In vivo , TLD-1A8A has the ability to increase the severity of adoptively transferred EAE. Thus, ASF is a molecule with a previously unsuspected immunological function. The data presented in this thesis indicate that ASF is a molecule expressed by antigen presenting cells whose function is to dampen T cell responses. This work provides a new understanding of a molecule involved in controlling immunological processes.
