Abstract	Cyclin-dependent kinases (CDKs) and their associated cyclins are key regulators of cell cycle progression. Caenorhabditis elegans has two predicted A- type cyclins, encoded by the genes cya-1 and cya-2. Cyclin A has been shown to be required for entry into mitosis in other organisms. However, the functions of C. elegans cyclin A remain to be determined. Here I present the characterization of a partial loss-of-function mutation of the cya-1 gene, bc416. cya-1 (bc416) causes cell division defects in certain cell lineages during embryogenesis, including in the NSM lineage. Furthermore, I found that CYA-1 promotes the transition from S to M phase in these lineages, which is consistent with the function of A-type cyclin in other systems. In addition, I present evidence that CDC-25.2, which is one of 4 cdc-25 phosphatases in C. elegans, activates the cyclin A/CDK complex to promote cell cycle progression. To my knowledge, this is the first demonstration of a functional interaction between the cyclin A/CDK complex and a Cdc25 phosphatase in C. elegans. The coordination of cell proliferation and cell fate determination is critical during development. However, the mechanisms through which this is accomplished are unclear. Previous studies have shown that the Snail-related protein CES-1 of C. elegans represents a functional link between asymmetric cell division and apoptosis specifically in the NSM lineage. CES-1 regulates cell polarity in the NSM neuroblast, and following asymmetric division of the NSM neuroblast, CES-1 is predominantly present in one daughter cell, the NSM. Here is suppresses apoptosis by repressing transcription of the pro-apoptotic BH3-only gene egl.-1. I demonstrate that CES-1 enhances the NSM neuroblast division defect caused by cya-1 (bc416). This result indicates that apart from regulating cell polarity and apoptosis, CES-1 affects cell cycle progression in the NSM lineage.
