Abstract	Epilepsy is associated with cognitive impairments which often manifest as a higher prevalence of memory impairments. Memory impairments in patients with epilepsy may persist even with sufficient control of seizures, suggesting other factors may contribute to cognitive outcomes. In this context spatial navigation may be used as a proxy for memory function. Although this is a highly reproducible result in preclinical models, the translation of these findings to humans remains difficult. Furthermore, how this information is consolidated during the subsequent sleep period remains unknown. Chapters 4 and 5 provide evidence that rats in the pilocarpine model of TLE show marked impairments in the Morris water maze task and are accompanied by severe cell loss in the CA1 layer of the hippocampus, a brain region thought to be critical for performance of spatial tasks. Chapter 4 shows that these impairments are not associated with time spent in or the oscillatory power during sleep in the preceding period. Finally, Chapter 5 outlines the study which shows that hippocampal replay in sleep, a potential memory consolidation mechanism, is intact in TLE rats. Chapter 6 presents evidence that humans with temporal lobe epilepsy (TLE) do not show impairments in a virtual water maze task. This chapter further shows the patients' performance is not related to the lateralization of the seizure foci nor the lateral dominance of memory function.
