Abstract	Swarming motility is a flagella-dependent surface motility, and is also a form of bacterial group behavior. Similar to its counterpart biofilm formation, our understanding of swarming motility has been hampered by its complex set of requirements including quorum sensing (QS) systems, surfactant production, flagella, and the second messenger molecule c-di-GMP. Previous work in our laboratory found that in response to a physiologically important amino acid - arginine - P. aeruginosa is repressed for swarming with a concomitant increase in intracellular c-di-GMP. My doctoral research has focused on elucidating mechanisms, both c-di-GMP-dependent and - independent, that regulate swarming motility on an arginine-containing medium. This work characterizes two mechanisms by which the extracellular arginine impacts swarming motility. The PQS (Pseudomonas Quinolone Signal) QS system, and in particular its intermediate product HHQ (2-heptyl-4-hydroxyquinoline) regulates phenazine production, which represses swarming motility in a c-di-GMP-independent manner. Meanwhile, an orphan response regulator - PA14_20780 - regulates swarming motility by either directly binding to or influencing flagellar proteins' function(s) or indirectly via modulating intracellular c-di-GMP levels. Furthermore, mutagenesis of the putative functional domains of PA14_20780 indicate that REC and ATPase domains are essential in regulating swarming motility in P. aeruginosa in response to arginine. Overall, this study contributes to our growing knowledge of bacterial swarming motility in P. aeruginosa. In addition to the identification of PQS/HHQ signals, and an orphan response regulator, we have constructed a comprehensive mutant library targeting various c-di-GMP metabolism proteins. This library will be useful for future investigations, be it swarming motility or other phenotypes in P. aeruginosa.
