Abstract	Eukaryotic cells are compartmentalized into membrane-enclosed organelles. Genetic screens and reconstituted biochemical assays have revealed that intracellular protein trafficking is mediated by a conserved mechanism of vesicle budding, transport, and fusion. Central to all intracellular membrane transport events is a conserved family of membrane proteins, termed SNAREs. In this study, I report my studies into the physical associations of yeast vacuole SNARE proteins. Through affinity purification and biochemical studies of tagged SNAREs, I have found that vacuole SNARE proteins interact as a complex with another multisubunit protein complex, termed the HOPS (Homotypic vacuole fusion and Protein Sorting) complex. HOPS associations with vacuole SNARE complexes are directly opposed by another SNARE regulatory protein, Sec17p (yeast ?-SNAP). Sec17p mediates SNARE complex binding and disassembly by the ATPase Sec18p (yeast NSF). In addition to its interactions with SNARE complexes and Ypt7p, we report that HOPS also binds directly to the phosphatidyl inositol 3-phosphate binding PX domain of the SNARE Vam7p. We suggest that through these interactions, HOPS regulates Ypt7p-dependent vacuole tethering, the transition to SNARE-mediated vacuole docking, and the recruitment of regulatory lipids and proteins to the vertex ring membrane microdomain where fusion occurs.
