Abstract	Sex determination in C. elegans leads to the development of sex-specific structures and contributes to the regulation of sexually dimorphic programmed cell death. A search for factors that regulate the sexually dimorphic cell death of the male-specific cephalic companion (CEM) neurons led to the identification of the novel gene tra-4 as a new general regulator of sex determination in C. elegans. tra-4 , which encodes a PLZF like zinc finger DNA-binding protein, is not only required for feminization but also represents new member of the synMuvB class of genes. Together with the synMuvA class of genes, the synMuvB class of genes antagonizes let-60 /Ras signaling during vulva development. A subgroup of the synMuvB class of genes, including lin-35 /Rb and Myb-complex-related genes, encodes a multi-protein repressor complex named Myb-MuvB. This complex is implicated in transcriptional repression through remodeling chromatin structure and/or modifications of histones. I demonstrate that the Myb-MuvB complex has a role in feminization. In contrast, most of the synMuvB genes that do not encode members of this complex have no role in feminization. In addition, I provide genetic evidence that TRA-4 is a component of the Myb-MuvB complex and demonstrate that tra-4 acts in parallel to the central sex determination pathway. This line of evidence suggests that TRA-4, possibly together with the Myb-MuvB complex, acts in parallel to the central sex determination pathway during hermaphrodite development. Moreover, the data I provide suggests that the TRA-4/Myb-MuvB complex acts in parallel, or together, with TRA-1/Ci/Gli to repress male-specific gene transcription in hermaphrodites through remodeling chromatin structure and/or modifying histones.
