Abstract	The essential QSR1 gene in Saccharomyces cerevisiae is a member of a highly conserved family of genes found in eukaryotic and archaebacterial cells. QSR1 was discovered by its synthetic lethal relationship with QCR6, the nuclear encoded gene for subunit 6 of the mitochondiral bc1 complex. Despite being a gene that is found in many species, the function of the QSR1 gene product and its homologues was largely unknown. The goal of this study is to characterize the QSR1 gene product in yeast in an effort to understand its function in all cells in which it is present. Additionally, information on the function of Qsr1p can be used to guide future work on Qcr6p in an effort to understand its new found role outside of respiration. This study made use of a combined biochemical and molecular genetic approach to understand the function of Qsr1p. Initially, antibodies against Qsr1p were used to identify its subcellular location on the large ribosomal subunit. The conservation of function between distantly related homologues of QSR1 was demonstrated by rescuing the lethal phenotype of a QSR1 null strain with the human or corn homologues. Biochemical analyses of QSR1 mutants were used to demonstrate a role for the Qsr1p in the joining of 40S and 60S ribosomal subunits. It was also determined that Qsr1p is important for the stability and functioning of 60S subunits in translational elongation assays. Identification of Qsr1p as the large ribosomal subunit protein L7 shows that Qsr1p is an exchangeable ribosomal protein. This suggests that it may play a role in translational regulation by inactivating 60S subunits in its absence. Studies involving the cytosolic protein Sqt1p indicate that it plays an important role in the assembly of Qsr1p onto the 60S subunit. This report documents the role that Qsr1p plays in translation and perhaps translational regulation. This function will undoubtedly be found to be common to the many homologues of QSR1. The studies presented here also detail a number of aspects of Qsr1p function that can be exploited in future studies of Qcr6p.
