Abstract	The Arabidopsis catalase gene family contains at least three genes, encoding subunits which combine in tetramers to form 7 detectable isozymes. Two of these genes are within 250 bp of each other at a single locus (CAT3/1) near the top of chromosome 1. while the third, CAT2, is near the bottom of chromosome 4. The genes are 70-72% identical at the level of nucleotide sequence and 75-84% identical and 87-94% similar, considering conservative substitutions at the level of amino acid sequence. Sequence and mapping analysis of the three Arabidopsis catalases revealed evidence of two duplication events early in the evolution of this gene family. Both the individual isozymes and the individual subunit mRNAs show distinct patterns of spatial (organ-specific) expression. A maximum of seven isozymes are detected in flowers and leaves and a minimum of two are seen in roots. Similarly, mRNA abundance of the three genes varies among organs and in situ hybridization analysis detected cell specific expression of all CAT genes in some tissues. The three catalase genes show differential expression at the RNA and protein levels which suggests the possibility of distinct roles, given their combined expression patterns. Although expression levels varied, no tissue or cell expressed one subunit to the exclusion of the others. A ΔCAT3/1 line and CAT1 antisense lines show no detectable phenotype as yet; therefore the CAT2 subunit may be sufficient for function. CAT2 as the fundamental catalase is supported by the fact that an isozyme containing the CAT2 subunit is present in all tissues examined. Therefore it may be necessary to eliminate CAT2 before generating a strong phenotype in Arabidopsis. Equally plausible is a level of functional redundancy that would require loss of each of the three CAT genes before an obvious phenotype becomes evident.
