Abstract	The docking and fusion of cellular membranes plays an essential role in the secretory and endocytic pathways and in organelle inheritance. The homotypic fusion of vacuoles from the yeast Saccharomyces cerevisiae has been used as a model system to study this fundamental and highly conserved process. Vacuole fusion occurs in stages of priming, docking, and fusion, and utilizes factors common to other cellular membrane fusion events: Sec18p (yeast NSF), Sec17p (yeast (α-SNAP), a Rab GTPase, and v- and t-SNAREs. The work in, this thesis demonstrates that Vam2p and Vam6p, which are conserved across species but lack structural homologs in other yeast trafficking reactions, are components of a large complex ("Vam2/6p") on the vacuole membrane which is required for docking. This complex interacts directly with the Rab-GTPase Ypt7p and is regulated by the membrane priming activity of Sec18p. Among the vacuole SNAREs, only the s-SNARE (SNAP-23/25 homolog) Vam7p is needed for the Sec18-dependent regulation of Vam2/6p. Strikingly, both Vam7p and Vam2/6p depend on Ypt7p and docking for their continued vacuole adherence, suggesting that they participate directly in vacuole:vacuole contact. Inhibition of Vam2p function prevents trans -SNARE pairing, and a small portion of the v- and t-SNAREs is complexed with Vam2/6p. Vam7p and Vam2/6p thus provide functional and physical links between membrane priming, docking, and trans -SNARE pairing.
