Abstract	The eukaryotic secretory pathway delivers proteins and lipids to their correct destination in a regulated manner. Transport through the secretory pathway is mediated by vesicles or tubular elements that bud from donor compartments. These transport intermediates then undergo site-specific tethering and fusion events with cognate acceptor compartments, allowing delivery of secretory cargo to their proper destinations. Biochemical, genetic and morphological studies on the yeast Saccharomyces cerevisiae have provided a wealth of information regarding vesicular transport between the ER and Golgi compartments. Much of the secretion machinery in yeast is conserved with that of higher organisms, making yeast an informative model organism in which to elucidate underlying mechanisms. In this dissertation, the function of the conserved integral membrane protein Yip1p in ER to Golgi transport is investigated. Biochemical and genetic inhibitors of Yip1p function block COPII-dependent budding from ER membranes in cell-free assays. Morphological and genetic experiments corroborate these findings, indicating Yip1p has a previously unappreciated role in COPII vesicle biogenesis. In addition, a genetic approach allowed the identification of YOS1 ( Y ip o ne s uppressor 1 ) as a novel multi-copy suppressor of the thermosensitive yip1-4 strain. YOS1 encodes a conserved integral membrane protein of 87 amino acids that forms a complex with Yip1p and Yif1p. This work indicates that a heterotrimeric Yip1p-Yif1p-Yos1p protein complex provides an essential function in the production of COPII vesicles from ER membranes.
